Each antibody clone is evaluated for target specificity, sensitivity, and reproducibility. Selection of suitable antibodies requires:

  • Binding of intended target
  • Differentiation of signal from background noise
  • Detection of low levels of antigen
  • Consistent results from similar samples

After selection of suitable antibody clones, antibody conjugates are evaluated for proper expression and localization:

  • Signal is specific
  • Counter stains validate specificity
  • Staining is distinct
  • Localization and cellular distribution are confirmed

Multiplexed panels of antibody clones are optimized to minimize the number of cycles needed. All antibodies in the final multiplex panel must show:

  • Distinct staining similar to singleplex
  • No steric hindrance or nonspecific cross-talk
  • Localization, cellular distribution, and percentage of target populations consistent with singleplex

Each tissue or cell sample used for validation is prepared carefully to ensure sample integrity. Multiplexing panels must pass all antibody validation criteria in:

  • At least three suitable tissue types
  • Each tissue tested with at least two replicates
